摘要
目的:动态观察大鼠C6胶质瘤模型的自然生长过程及反义IGF-1mRNA对胶质瘤的治疗效果和治疗机制。方法:将32只Wistar大鼠分成4组,每组8只,(1)对照组(C组)于右尾状核接种C6鼠胶质瘤细胞。(2)治疗组(T组):于右尾状核接种C6细胞后的第7、10天在接种点注射脂质体包裹的质粒P-anti-IGF-1,对胶质瘤进行治疗于右尾状核接种经反义IGF-1mRNA转染的C6胶质瘤细胞。
To study the value Of 1.5 T MR in the long-term follow-up of rat C6 glioma model and the effects of anti sense IGF- 1 mRNA on tumor tumor growth in vivo. Methods:Thirty-two Wistar rats were diveded into four groups, eight rats for each group: (l)Group C was control group, rats were inoculated with C6 cells in right caudate nucleus. (2)Group T was ex- perimental group, the antisense mRNA were injected into tumol mass on day 7 and day 10 after implantation of C6 cells. (3) Group K,C6 cells transfected by antisense IGF-l mRNA in vitro (K cell for short) were implanted into caudate nucleus. (4) Group KC, injected C6 cells to the animal in which tumor in right hind leg induced by K cells had just regressed up for 2 weeks. All animals were examined by plain and enhancement MR scanning at l, 2, 4, 8weeks after implantaion. One rat in each group was sacrificed at every time point for pathology examination. Monoclonal antibody to CD4 and CD8 lymphocytes were used to detect the host immune response aroud tumor with immunohistochemical staining. Results:The growth of rat C6 glioma model and its changes after treatment could be shown clearly on 1. 5T MR imaging. The tumor growth was sig- nificantly inhibied in group T compared with group C. The host immune response were detected in groups T,K and KC.
