摘要
目的:优化梯度离心法提取人血小板过程中富含血小板血浆(Platelet rich plasma,PRP)的分离条件,以提高血小板的获得率及质量。方法:梯度离心分离人血小板,其中初次离心采用正交实验设计,以离心力和离心时间为实验因素,均设置五水平,获得富含血小板血浆,利用血细胞分析仪分别计数离心前全血及离心后PRP中血小板数量,计算血小板获得率,并且涂片计数PRP中的杂质细胞。结果:离心力和离心时间对提取PRP中血小板效率和纯度有显著影响,并且两者存在交互作用。结论:采用离心力为100 g,离心时间为15 min分离提取的PRP中血小板数量较多,纯度较高。
Objective:To optimize the centrifugal separation of PRP for improving the isolation of platelets from human blood.Methods:In the separation of PRP,the first step of platelet isolation,centrifugal force and time were considered main factors,both at five levels through orthogonal experiment design.Platelets of blood and PRP were counted individually with hematology analyzer to calculate the platelet pick-up rate of the first step.WBC and RBC were detected in PRP smear under microscope to provide the information of isolation quality.Results:Centrifugal force and time were confirmed to be major factors in centrifugal separation of PRP,with interacting between each other.Conclusion:The centrifugal force at 100gravity and centrifugal time at 15minutes are expected to be the optimized condition of the centrifugal separation of PRP in platelets extraction from human blood.
