摘要
目的探讨细胞膜P-糖蛋白(P-gp)在人脑胶质瘤组织中的表达及原代胶质瘤细胞P-gp蛋白阳性表达与长春新碱(VCR)的敏感性的相关性。方法采用免疫组化方法检测37例人脑胶质瘤P-gp蛋白的表达;人脑胶质瘤细胞原代培养,四甲基偶氮唑蓝(MTT)比色法检测VCR药物的敏感性。结果 37例脑胶质瘤P-gp阳性表达率为54.1%(20/37),P-gp在Ⅱ~Ⅳ级脑胶质瘤的阳性表达率分别为39.4%、66.7%和87.5%。不同病理级别的胶质瘤患者P-gp阳性表达率间差异有统计学意义(χ2=8.529,P<0.05),病理级别与P-gp的表达强度呈正相关(r=0.479,P<0.05)。P-gp阳性表达与胶质瘤细胞对VCR的耐药有相关性(r=0.429,P<0.05)。结论 P-gp的表达与脑胶质瘤恶性程度及其对VCR的耐药性相关,检测P-gp的表达及其对VCR的敏感有助于指导脑胶质瘤的个体化治疗。
Objective To explore the P-gp protein expression in human glioma and its correlation with primary glioma cells′ drug sensitivity to vincristine. Methods Immunohistochemical method was used for the detection of human glioma P-gp expression in 37 cases.Primary human glioma cell was cultured,and assay vincristine(VCR) drug sensitivity was tested by methyl thiazolyl tetrazolium method(MTT). Results Of this 37 cases,positive expression of the glioma P-gp rate was 54.1%(20/37),and the rates in glioma from grade Ⅱ to Ⅳ were respectively 39.4%,66.7%,and 87.5%.Patients of different pathological grades had significantly different P-gp expressions(χ2=8.529,P0.05),and pathological grades were positively correlated to P-gp protein expression(r=0.479,P0.05).P-gp expression was relevant to glioma cells′ drug-resistance against vincristine(r=0.429,P0.05). Conclusion P-gp expression is relevant to the malignance of glioma tumors as well as the drug-resistance against vincristine.A combined detection of P-gp expression and vincristine drug sensitivity is of great value for instructing individualized therapy for glioma sufferers.
