摘要
目的 :为了解糖尿病早期肾小管和集合管上皮细胞凋亡过程中ICE、Bcl 2、Bax、p5 3是否参与调控。方法 :雄性Wistar大鼠 6 0只 ,四氧嘧啶 (alloxan)尾静脉注射 (5 0mg/kg)造糖尿病模型 ;石蜡包埋肾切片经ICE、Bcl 2、Bax、p5 3免疫组化染色。结果 :ICE、Bcl 2、Bax、p5 3蛋白分别在肾小管和集合管上皮细胞两次凋亡发生前或发生时表达。结论 :p5 3蛋白的表达可能反应了高血糖致使肾小管、集合管上皮细胞DNA损伤 ;Bax表达增强可能有使Bcl 2与Bax的比值发生变化 ,以及在两次凋亡发生前ICE表达增强均与受损的肾小管、集合管上皮细胞凋亡有关。
Objective:The aim of this study was to understand the roles of ICE、Bcl 2、Bax and p53 in the process of apoptosis in the tubular and collecting tubules epithelial cells of rats with diabetes mellitus (DN)for 140 days. Methods:60 male wistar rats were injected with alloxan(50mg/kg)by tail vein;immunohistochemical techniques were used to identify ICE、Bcl 2、Bax and p53 showing mmunoreaction in the different sections of kidneys. Results:ICE、Bcl 2、Bax and p53 expressed in the opithelial cells of the proximal tubules, the medullary loop and the distal tubules before or on the apoptosis, respectively. Conclusion:Expression of p53 implied that it resulted in the DNA damage of the tubular and collecting epithelial cells. The expression of Bax altered the ratio of Bcl 2 to Bax, and the expression of ICE accelerated before two apoptotic emergence, they were probably related to the apoptosis of the injured tubular and collecting epithelial cells.
