王浆蛋白质及其生理药理作用分析

Analysis of royal jelly proteome and its physiological and pharmacological function

目的从蛋白质组的角度分析王浆及其生理药理作用,为进一步理解王浆医用价值提供理论基础。方法提取王浆全蛋白质组分进行双向电泳(2-DE)和质谱鉴定(MALDI-TOF/MS),然后利用蛋白质数据库进行比对。结果 2-DE图谱共检测出152个蛋白点,对其中57个高丰度蛋白进行了MALDI-TOF/MS鉴定,有45个属于王浆主蛋白(MRJPs)家族,还有3个葡萄糖氧化酶,1个过氧化物还原酶,1个谷胱甘肽S转移酶,其余7个蛋白点未能找出匹配蛋白。结论这些蛋白质和酶类具有营养、免疫调节、抗氧化、抗炎、抗肿瘤等多种生物活性,或许可以部分说明王浆对于人类的医药保健作用。

ObjectiveTo make a proteomic analysis of royal jelly（RJ） and its physiological and pharmacological function and to make a further understand of RJ＇s medical values.MethodsRJ protein complement was extracted and experienced two dimensional electrophoresis（2-DE） and matrix-assisted laser desorption ionization-time of flight mass spectrometry（MALDI-TOF/MS）.Then a protein engine identification tool was applied to protein alignment.ResultsOne hundred and fifty-two proteins were detected in 2-DE,of which 57 high abundant proteins were identified.Forty-five were assigned to major royal jelly proteins（6 MRJP1,11 MRJP2,20 MRJP3,4 MRJP4,4 MRJP5）,three were glucose oxidase（GOX）,one was peroxiredoxin（PRDX）,one was glutathione S-transferase（GST S1）,and seven were not identified due to a deficiency of protein.ConclusionThese proteins and enzymes have several biological activities,such as nutrition,immunoregulation,protection against oxidation,anti-inflammatory and antitumor,which may partly explain RJ＇s pharmacological traits for human beings.
