摘要
目的:建立龙胆科白族药药材的龙胆苦苷的含量测定法。方法:采用色谱:Waters C18(150mm×4.6mm,5μm);以甲醇-0.05%磷酸作线性梯度洗脱;流速为0.6mL/min;检测波长为254nm;柱温为30℃。结果:6种药材中龙胆苦苷的色谱峰与共存组分完全达到基线分离,线性范围0.0575~2.3μg(r=0.9999),平均回收率为97.71(RSD=1.38)。结论:所建立的方法测定快速,结果准确可靠。
Aim: To establish a RP-HPLC method for the determination of Gentiopicrin in six gentianaceae herbs.Methods:The samples were separated on a Waters Symmetry C18(4.6mm×150mm,5μm) with the mobilephase consisting of methanol and water(containing 0.05 %phosphoric acid) at a flow rate of 0.6 ml/min.The operation was conducted in the gradient mode in which the ratio of methanol to water varied from 20∶80 to 30∶70 in 30 min.The detecting wavelength and the temperature of column were at 254 nm and at 30℃ respectively.Results:Chromatographic separation of gentiopicin and the coexisted compounds achievedbaseline resolution.The calibration was linear in the range of 0.0575~2.3μg(r = 0.9999),and the average recoverywas 97.71(RSD = 1.38).Conclusion:The method is rapid and precise.
