摘要
目的研究高三尖杉酯碱和异三尖杉酯碱对人早幼粒白血病细胞HL-60凋亡的影响。方法采用流式细胞术、DNA凝胶电泳、光镜及透射电镜等方法。结果证明高三尖杉酯城及异三尖杉酯碱能快速、显著地诱导HL-60细胞的凋亡,其作用有明显的浓度及时间依赖性。高三尖杉酯碱的活性明显高于异三尖杉酯碱。结论高三尖杉酯碱及异三尖杉酯碱皆可诱导HL-60细胞凋亡,但高三尖杉酯碱的生物活性明显高于异三尖杉酯碱。
To study the effects of homoharringtonine and isoharringtonine on the induction of apoptosis on human promyelocytic leukemia HL-60 cells. Methods By using flowcytometry,DNA gel electrophoresis, microscopy and electron microscopy. Results it wasdemonstrated that homoharringtonine and isoharringtonine can induce apoptosis of HL-60cells significantly and rapidly. The activity of homoharringtonine was higher than that of isoharringtonine,and it was time and concentration dependent. Conclusions Both homoharringtonine and isoharringtonine could induce apoptosis of HL-60 cells in terms of morphology,and DNA ladder assay. The activity of homoharringtonine was much higher than that ofisoharringtonine.
