蒿芩清胆汤及其化裁方对病毒性肺炎湿热证小鼠炎性细胞因子的影响 被引量:8

Effect of Haoqin Qingdan Decoction and Its Modified Prescriptions onInflammatory Factors in Viral Pneumonia Mice with Damp-heat Syndrome

摘要 【目的】观察蒿芩清胆汤及其化裁方对病毒性肺炎湿热证小鼠炎性细胞因子肿瘤坏死因子(TNF-α)、白细胞介素-6(IL-6)表达及肺组织流感病毒含量的影响。【方法】选用BALB/c小鼠,随机分为正常对照组、湿热证模型组、蒿芩清胆汤组(剂量为36.92 mg.kg-1.d-1)、化裁1组(剂量为30.39 mg.kg-1.d-1)、化裁2组(剂量为18.44 mg.kg-1.d-1)、化裁3组(剂量为32.73 mg.kg-1.d-1)、化裁4组(剂量为29.09 mg.kg-1.d-1);除正常对照组外,其他各组均采用高脂饲料+气候箱暴露(10 d)法复制湿热证模型,第11天接种H1N1病毒;取各组小鼠肺组织采用荧光定量聚合酶链反应法检测流感病毒mRNA、TNF-αmRNA、IL-6 mRNA表达水平。【结果】蒿芩清胆汤及各化裁方组均可降低肺组织流感病毒mRNA表达水平,与湿热证模型组比较差异有显著性意义(P<0.05或P<0.01);湿热证模型组肺组织TNF-αmRNA、IL-6 mRNA表达水平升高,与正常对照组比较差异具有显著性意义(P<0.01),各治疗组均可不同程度降低2种炎性细胞因子的表达水平,其中蒿芩清胆汤组,化裁2组(全方减竹茹、法夏、滑石、茯苓、甘草),化裁3组(全方减枳壳、陈皮)与湿热证模型组比较差异有显著性意义(P<0.05或P<0.01),而化裁1组(全方减青蒿、黄芩),化裁4组(全方减大青叶、板蓝根)与湿热模型组比较差异无显著性意义(P>0.05)。【结论】蒿芩清胆汤治疗流感病毒性肺炎湿热证的作用与其能抑制流感病毒复制,下调炎性细胞因子TNF-α、IL-6的基因表达水平有关,拆方研究结果表明蒿芩清胆汤全方效果最佳。 Objective To investigate the effect of Haoqin Qingdan Decoction(HQD)and its modified prescriptions on mRNA expression of inflammatory factors of tumor necrosis factor alpha(TNF-α)and interleukin 6(IL-6)as well as influenza virus in the lung tissue of viral pneumonia mice with damp-heat syndrome.Methods BALB/c mice were randomized into the normal control group,model group,HQD group(in the dose of 36.92 mg·kg-1·d-1),modified HQD group 1(in the dose of 30.39 mg·kg-1·d-1),modified HQD group 2(in the dose of 18.44 mg·kg-1·d-1),modified HQD group 3(in the dose of 32.73 mg·kg-1·d-1),and modified HQD group 4(in the dose of 29.09 mg·kg-1·d-1).Except for the normal control group,the mice in other groups were fed with high-fat forage and were put into the climate box for 10 days to induce damp-heat syndrome,and then were inoculated with H1N1 virus on the 11th day.Fluorescence quantitative polymerase chain reaction(PCR)was used for the detection of mRNA expression of TNF-α,IL-6 and influenza virus in mice lung tissue of different groups.Results HQD and its modified prescriptions had an inhibition on mRNA expression of influenza virus as compared with the model group(P〈0.05 or P〈0.01).TNF-α and IL-6 mRNA expression levels were higher in the model group than those in the normal control group(P〈0.01).HQD group,modified HQD group 2(HQD with Caulis Bambusae in Taenia,Rhizoma Pinelliae,Talcum,Poria,Radix Glycyrrhizae removed),and modified HQD group 3(HQD with Fructus Aurantii and Pericarpium Citri Reticulatae removed)had lower TNF-α and IL-6 mRNA expression levels than the model group(P〈0.05 or P〈0.01).TNF-α and IL-6 mRNA expression levels in modified HQD group 1(HQD with Herba Artemisiae Annuae and Radix Scutellariae removed)and modified HQD group 4(HQD with Folium Isatidis and Radix Isatidis removed)did not differ from those in the model group(P〉0.05).Conclusion The therapeutic mechanism of HQD for viral pneumonia mice with damp-heat syndrome is related with the inhibition of influenza virus replication and with the down-regulation of inflammatory factors of TNF-α and IL-6 mRNA expression.HQD has a better effect than its modified prescriptions.
