摘要
建立了基于自制简易固相萃取柱和高效液相色谱联用测定食品中五种合成着色剂的新方法。样品提取液经聚酰胺粉吸附后,将聚酰胺粉装填到25 mL塑料注射器针筒内制成简易固相萃取柱;然后用甲醇-甲酸去除杂质,再用乙醇―氨水―水混合液洗脱合成着色剂;最后将洗脱液经乙酸中和,蒸干,加水定容,待测。本研究同时优化了固相萃取的条件,在有效去除杂质的同时获得了很高的回收率。本研究有效的解决了国标方法中操作繁琐,前处理耗时长,检测通量低的弊端;利用聚酰胺固相萃取柱大大提高了检测效率,能够很好的满足食品检测机构检品数量大的要求。
Based on the joint use of the simply self-made Solid Phase Extraction Column and high performance liquid chromatography(HPLC),a new method for determination of five kinds of synthetic colorants in foods was established.Rida extraction solution was processed by polyamide adsorption,and then the simple Solid Phase Extraction Column was made by adding the polyamide into the 25 mL syringe;then using the mixture methyl alcohol-formic acid to clear from the impurities,using the ethanol-ammonia-water to elute the synthetic colorants,and then adding acetic acid into the eluant,drying,volume filter,under the test.This study optimized the solid phase extraction conditions,not only can clear from impurities effectively,but also can get a high recovery rate.This study overcame the disadvantages in the international criterion methods,such as complex operation,long-time pretreatment and low throughout.Taking advantages of the polyamide Solid Phase Extraction Column,the detection efficiency was improved greatly,which can fulfill the quality demand of the Food Testing Administration.
