摘要
建立苯巴比妥血清药物浓度的测定方法,用RP-HPLC法检测,Nova-PaK C18柱,流动相:甲醇-KH2PO4(0.01 mol/L,pH=3.50)(50∶50);柱温:25℃;流量:1 mL/min;检测波长:311 nm。苯巴比妥、苯妥英钠与卡马西平线性范围分别为5~40μg/mL,5~40μg/mL和2.5~20μg/mL,平均回收率分别为99.95%9,9.36%和99.65%日内、日间RSD均小于5%,标准曲线相关性良好。该法简便、经济,结果准确,适合临床应用。
The method of determination of serum drug concentration in phenobarbital was established.RP-HPLC was adopted for the determination.By this method,the linear ranges of PB,DPH and CBZ were 1.6~12.8 μg/mL,1.6~12.8 μg/mL and 0.8~6.4 μg/mL,respectively.The average recovery rate of PB and CBZ were 99.95% and 99.65%,respectively.The relative standard deviations(RSD) of within a day and inter-day were less than 5%.The calibration curve presented good linearity.This method was simple,economical,accurate and sensitive,and suitable for clinical application.
