摘要
目的通过动物实验研究消痰散结方及其拆方(消痰方、解毒方、散结方)对S180荷瘤鼠肿瘤生长的影响,及对肿瘤相关基因蛋白表达的影响;探讨消痰散结方及其拆方的作用效果。方法皮下种植S180肿瘤细胞,制备荷瘤鼠,分为4组,分别给予中药干预包括消痰散结方及其拆方(消痰方、解毒方、散结方),至3周处死小鼠,称瘤重,MaxVision二步法检测肿瘤相关血管内皮生长因子(VEGF)、激酶插入区域受体(KDR)、E钙黏附蛋白、环氧化酶(COX)-2、端粒酶、细胞间黏附分子(ICAM)-1,比较4种中药干预后表达的差异。结果 VEGF、KDR、COX-2、端粒酶、ICAM-1均呈现消痰散结方组有表达降低趋势,E-Cad表达升高趋势,经分析消痰散结方与消痰方比较VEGF、ICAM-1表达差异有统计学意义(P<0.05),消痰散结方与解毒方比较KDR表达差异有统计学意义(P<0.05)。结论消痰散结方作为消痰方、解毒方、散结方的合方,在肿瘤治疗上优于其拆方。
Objective To study the effects of Xiaotan Sanjie recipe and its decomposed recipes (Xiaotan recipe, Jiedu recipe, Sanjie recipe) on S180 tumor growth and associated gene protein expression, and to explore the efficacy of Xiaotan Sanjie recipe and its decomposed recipes. Methods Subcutaneous implantation of S180 tumor cells was performed to establish tumor-bearing mice models which were then divided into 4 groups to be treated with Xiaotan Sanjie recipe and its decomposition recipes (Xiaotan recipe, Jiedu recipe, Sanjie recipe). After 3 weeks, the mice were killed, and the tumors were weighted. Max Vision system was used to detect expressions of tumor-associated vascular endothelial growth factor (VEGF), kinase domain receptor (KDR), E-Cadherin, cyclooxygenase-2 (COX-2), telomerase and immune cell adhesion molecule -1 (ICAM-1) after intervention with the four recipes. Results Xiaotan Sanjie recipe group showed reduced expressions of VEGF, KDR, COX-2, telomerase and ICAM-1, and increased expression of E-Cadherin. In Xiaotan Sanjie recipe group, the expressions of VEGF and ICAM-1 in were significantly different with those in Xiaotan recipe group (P0.05), as was expression of KDR with that in Jiedu recipe group (P0.05). Conclusion Xiaotan Sanjie recipe, as an integration of Xiaotan, Jiedu and Sanjie recipes, may be more effective in treatment of tumor than any of these decomposed recipes alone.
