摘要
目的探讨吡格列酮对糖尿病大鼠胰岛β细胞的保护作用。方法将雄性Wistar大鼠30只随机抽取10只作为空白对照组,其余腹腔注射链脲佐菌素(STZ)建立糖尿病大鼠模型。将成模大鼠(20只)随机分为糖尿病对照组和吡格列酮组。吡格列酮组给予吡格列酮10mg·kg-1·d-1灌胃,其余2组给予等量生理盐水灌胃。16周后3组大鼠断尾测空腹血糖(FPG),心包取血测空腹胰岛素水平(FINS)和糖化血红蛋白(HbAlc)。观察3组大鼠FPG、FINS、HbAlc、胰岛素抵抗指数(HOMA-IR)水平并比较。结果与糖尿病对照组相比,吡格列酮组FPG、HbAlc、FINS、HOMA-IR水平降低(P<0.05);糖尿病对照组、吡格列酮组FPG、HbAlc、FINS、HOMA-IR水平较空白对照组升高(P<0.05)。结论吡格列酮对胰岛β细胞有一定的保护作用。
Objective To discuss the protective effect of pioglitazone on pancreatic to islet β cell in diabetes mellitus rats.Methods 30 healthy male Wistar rats were enrolled,10 rats were randomly assigned to normal control group,and the others were intraperitoneally injected with STZ to establish diabetes mellitus(DM) rat model.After successful modeling,the rats were assigned to DM control group and pioglitazone group.Pioglitazone group were given pioglitazone 10 mg·kg-1·d-1 intragastric administration.The other two groups were given sodium chloride.After sixteen weeks,mearsured and compared the fasting blood glucose(FPG),fasting insulin(FINS),glycosylated hemoglobin(HbAlc) and insulin resistance index(HOMA-IR) of 3 groups.Results The levels of FPG,HbAlc,FINS,HOMA-IR in pioglitazone group were lower than those in DM control group(P0.05).The levels of FPG,HbAlc,FINS,HOMA-IR in DM control group and pioglitazone group were higher than those in normal control group(P0.05).Conclusion Pancreatic to islet β cell has some protective effect.
