大肠杆菌分泌型载体表达重组人碱性成纤维细胞生长因子

SECRETION OF RECOMBINANT HUMAN bFGF EXPRESSED BY E.coli

设计构建了带有大肠杆菌外膜蛋白（Ｏｍ ｐＡ）信号肽序列的重组分泌型表达载体ｐＳＥ－ｈｂＦＧＦ，在大肠杆菌ＤＨ５α中分泌表达了重组人碱性成纤维细胞生长因子（ｒｈｂＦＧＦ）。ＳＤＳ－ＰＡＧＥ、免疫印迹及生物活性测定均表明表达产物分泌到细菌细胞周质和培养基中。分泌到周质中的ｒｈｂＦＧＦ可占周质总蛋白的１５％ 。

Human basic fibroblast growth factor (hbFGF) gene was inserted into expression vector with OmpA signal sequence and expressed in E.coli DH5α. SDS PAGE analysis, western blot and bioassay verified that the expressed rhbFGF was secreted into culture medium and periplast of bacteria. The yield of rhbFGF secreting into periplast accounted for 15% of total periplast protein.
