摘要
目的探讨P选择素和细胞间粘附分子-1(ICAM-1)在肾缺血再灌注损伤中的作用。方法建立肾缺血再灌注损伤大鼠模型,观察缺血再灌注后肾组织P选择素和ICAM-1的表达,并用P选择素单克隆抗体进行了治疗。结果发现缺血再灌注后肾小管上皮细胞空泡变性和片状坏死,血尿素氮(BUN)和肌酐(SCr)水平升高;但再灌注前5min经P选择素单抗处理的动物肾组织与正常相近,肾小管上皮细胞未见变性及坏死,血BUN和SCr水平明显减低。免疫组化检测提示再灌注后好组织P选择素和ICAM-1表达增强,单抗治疗组肾组织未见P选择素和ICAM-1表达。结论P选择素和ICAM-1与肾缺血再灌注损伤相关,P选择素单抗对此具有保护作用。
Objective To investigate the role of P- selectin and intercellular adhesionmolecule - 1 (ICAM - 1) in renal ischemia/reperfusion injury. methods Adhesion moleculeexpression was measured by immunohistochemistry and renal function was detected in ratswith renal (I/R) injury. Some I/R rats were treated with anti - P - selectin monoclonalantibody (mAb). Results UP-regulated expression of P - selectin and ICAM - 1 in renaltissue, renal histological lesions and renal failure were present in the rats with I/R injury,while these changes were mild in the mAb treated group. Conclusion The results suggestedthat P - selectin and ICAM - 1 alight contribute to renal I/R injury, and anti - P - selectinmAb might prevent the kidney from I/R injury.
