摘要
从活性污泥中筛选出具备较高絮凝活性的菌株KJ-10。对菌株KJ-10原生质体的制备和再生的影响因素进行了考察。结果表明,KJ-10原生质体制备和再生的优化条件为:以1%接种量在CM液体培养基中活化12 h,酶解前加入5 mol.L-1甘氨酸预处理12 h,酶浓度为0.1 mg.mL-1,37℃水浴酶解120 min,并涂布于以0.5 mol.L-1蔗糖为稳定剂的再生平板。原生质体的形成率与再生率分别达到93.5%、20.5%。
The strain KJ-10 screened from the activated sludge had high flocculation activity.The influential factors for production and regeneration of flocculant-producing bacterium KJ-10 protoplast were investigated.The optimum conditions for production and regeneration of KJ-10 protoplast were determined as follows: activation for 12 h with 1% inoculation volume of liquid medium CM;pretreating with 5 mol·L-1 glycine for 12 h before enzymatic hydrolysis;enzymatic hydrolysis for 120 min in 37℃ water bath with enzyme concentration 0.1 mg·mL-1;coating on a renewable flat with 0.5 mol·L-1 of sucrose as stabilizer.The formation rate and regeneration rate of protoplast reached 93.5%,20.5%,respectively.
