摘要
以玉米杂交组合HiIIA×昌7-2和自交系"6308"为材料,采用体细胞筛选法进行抗草甘膦突变体的筛选,体细胞筛选采用了直接筛选和梯度筛选两种方式。结果表明,草甘膦最高筛选压力为0.5 mL/L,梯度筛选更有利于愈伤组织的存活和幼苗分化,同一筛选条件下,杂交组合HiIIA×昌7-2的分化时间明显短于自交系"6308",缓苗成活率和移栽苗成活率明显高于自交系"6308"。最终筛选得到在细胞水平上能抗0.5 mL/L草甘膦、植株水平上抗0.9 mL/L草甘膦的突变体。
Hybrid combination HiIIA×Chang7-2 and inbred line "6308" were chosen to select glyphosate resistant mutants in maize by the method of somatic screening in the experiment.Two kinds of screening methods such as direct screening and gradient screening were adopted in somatic screening.The results of preliminary study showed that the upmost selection pressure of glyphosate was 0.5mL/L.Gradient screening was better for survival of calli and differentiation of young plants.Combination hybrid HiIIA×Chang7-2 had obvious shorter differentiation time and higher survival rate not only in the nutrition pot but also in the field than inbred line "6308" under the same conditions of screening.As a result,the mutants which could resist 0.5 mL/L glyphosate at cell level and 0.9 mL/L glyphosate at plant level were obtained.
