摘要
目的 比较四环素类抗生素对全血吞噬细胞产生氧自由基(ROS) 和非细胞体系产生O·2 和·OH 的影响。方法 应用化学发光方法测定人全血吞噬细胞在受调理酵母多糖诱导下产生的ROS,黄嘌呤黄嘌呤氧化酶体系产生的O·2 和Vit CCuSO4酵母多糖H2O2 体系产生的·OH。结果 四环素类药物均有清除和抑制中性粒细胞产生氧自由基和O·2 的作用,但美满霉素作用最强,美满霉素还具有清除·OH 的作用,而其它四环素类药物无此作用。结论 四环素类药物可清除或抑制吞噬细胞产生的ROS和清除O·2 。
AIM To study the effects of tetracyclines drugs including tetracycline, minocycline, doxycycline and oxytetracycline on ROS generated by leukocyte and O · 2 ,·OH generated by cell free system. METHODS Chemiluminescence (CL) method was used to measure: ① reactive oxygen species (ROS) generated in whole blood leukocytes induced by opsonized zymosan; ② superoxide ( O · 2 ) induced by xanthine xanthine oxidase system; ③ hydroxy radicals (·OH) generated by Vit C CuSO 4 zymosan system. RUSULTS All drugs of tetracyclines had effects on inhibiting or scavenging ROS generated in leukocyte and O · 2 generated in the cell free system. The effect of minocycline is the strongest of all drugs. Minocycline had an effect on scavenging·OH, but other tetracyclines had not. CONCLUSIONS The all tetracyclines have effects on inhibiting or scavenging ROS generated by leukobytes and O · 2 generated by cell free system.
