Study on the relationship between apolipoprotein A5 gene-1131T/C polymorphism and homocysteinemia levels in patients with diabetic nephropathy

摘要 目的探讨载脂蛋白A5基因(APOA5)-1131T/C基因多态性与糖尿病肾病患者同型半胱氨酸(Hcy)水平的相关性。方法用聚合酶链反应-限制性片段长度多态性分析(PCR-RFLP)技术检测40例健康人及45例糖尿病肾病患者APOA5-1131T/C基因型,并统计等位基因频率分布情况;用Olympus2700型全自动生化分析仪检测同型半胱氨酸水平。结果糖尿病肾病组APOA5-1131T/C基因频率、等位基因频率与对照组差异无统计学意义(P>0.05)。糖尿病肾病组C等位基因携带者同型半胱氨酸水平显著高于非C等位基因携带者(P<0.05)。结论 APOA5-1131T/C基因多态性影响糖尿病肾病患者同型半胱氨酸水平,C等位基因与糖尿病肾病同型半胱氨酸水平增高有密切关系。 Objective To investigate the relationship between apolipoprotein A5 gene-1131T/C polymorphism and homocysteinemia levels in patients with diabetic nephropathy(DN).Methods The frequencies of genotypes and alleles of APOA5-1131T/C were detected in 45 DN patients by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP),and 40 healthy subjects were enrolled as controls.The levels of homocysteinemia in both groups were measured by Olympus AU 2700 analyzer.Results There was no significant difference in the frequencies of APOA5-1131T/C genotypes and alleles between DN patients and healthy subjects(P0.05).In DN group,the levels of homocysteinemia in C carriers(TC+CC) were significantly higher than those of non-C carriers(TT)(P0.05).Conclusion APOA5-1131T/C genepolymorphism is associated with the levels of homocysteinemia in DN patients.APOA5-1131T/C alleles may lead to the increase of homocysteinemia in DN.
