摘要
目的:观察家蚕细胞基因工程α1 干扰素(rhIFNα1) 对小鼠的免疫调节作用。方法:采用测定环磷酰胺30mg/kg 免疫抑制小鼠碳粒廓清速率,血清溶血素的方法观察rhIFNα10 .1 ,0 .4 ,1 .2 ×106IU/kg 对校正廓清指数,脾脏和胸腺重量以及溶血血红蛋白值的影响;利用离体胸腺细胞与rhIFNα115000 ,1500 ,150IU/ml 共孵,观察(rhIFNα1)对胸腺细胞的增殖作用。结果:rhIFNα1 增强免疫抑制小鼠巨噬细胞对碳粒的吞噬作用,使廓清指数增加( P<0 .01) ,脾脏重量增加( P< 0 .01) ,血红蛋白值有所增加;但rhIFNα11 .2 ×106IU/kg 却能使正常小鼠的血红蛋白值大幅度增加;rhIFNα11500 ,15000IU/ml 能促使胸腺细胞增殖,且15000IU/ml 能促进0 .1 % PHA 的增殖作用( P<0 .05) 。结论:家蚕细胞基因工程人α1
OBJECTIVE:To observe the immunity effects rhIFN α 1(recombined interferon α 1 using with genes of Chinese silkworm) on mice.METHOD:Measuring the serum phagocytic index,weight of the immune organ(spleen and thymus)and hemolytic hemoglobin of rhIFN α 1 0.1,0.4 and 1.2×10 6IU/kg in immunatory inhibited mice with cyclophaphamide(30mg/kg).Observe the proliferation effects of rhIFN α 1 by incubation of thymus cells with rhIFN α 1 of 15000,1500 and 150IU/ml.RESULTS:rhIFN α 1 enhanced the phagocytosis to carbon in immunatory inhibited mice,and increased the serum phagocyte index(P<0.01);it also increased the weight of spleen(P<0.05) and the hemolytic hemoglobin value.rhIFN α 11.2×10 6IU/kg improved the hemolytic hemoglobin greatly in normal mice;rhIFN α 11500,15000IU/ml accelerated the proliferation of lymphocyte in mice,and rhIFN α 115000IU/ml enhanced the proliferation effects of 0.1%PHa(P<0.05).CONCLUSION:rhIFN α 1 showed the immunatory improvement in mice at different aspects
