胎鼠宫内缺血缺氧后脑组织Bcl-2、Bax基因表达的研究

摘要 目的:观察胎鼠宫内缺血缺氧后脑组织Bcl-2、Bax 表达的动态变化。方法:钳夹一侧供应子宫的血管10m in 造成宫内缺血缺氧模型,分别再灌注0,1,4,8,15,24 h,进行半定量RT-PCR检测Bcl-2与Baxm RNA 的表达。结果:Bax 的表达在缺血缺氧当时即开始增强, 4 h 已有显著性(P< 0.01), 8 h 达高峰, 15 h 有所降低, 24 h 又出现升高( P< 0.01);而Bcl-2的变化无显著性(P> 0.05);Bcl-2与Bax 的比值在8 h 达最低。结论:在轻度宫内缺血缺氧后4 h,Bax Objecive:We examined the expression of apoptosis related genes, Bcl 2 and Bax, in intrauterus cerebral ischemia and hypoxia of rats.Methods: Ischemic hypoxic brain model was made in fetal rats by clamping the vessels supplied to one side corn of uterus. The vessels to the other side was used as sham control. Then observed fetal brains at reperfusion for 0, 1,4,8,15 and 24 hours after clamping 10 minutes. Semiquantitative reverse transcription polymerase chain reaction was used to reveal the expression of Bcl 2 mRNA and Bax mRMA. Results:Bcl 2 mRNA increased gradualy during reperfusion periods, but there were no significant changes (P>0.05). Bax mRNA increased unobviously after clamping 10 minutes, (P>0.05), but after 4 hours of reperfusion, it presented significant change (P<0.01), reached the peak level at about 8 hours, following a decreasing, it increased again at about 24 hours of reperfusin. The ratioes of Bcl 2/ Bax reached the lowest level during 8 hours. Conclusion:After mild hypoxia and ischemia, the changes of apoptosis rehated genes were already onset perhaps at about 4 hours of reperfusion, which provided the proof and also the clue for early treatment in newborns of intrauterus fetal distress.
