The Expression and Clinical Significance of RABEX-5 and RAB5 in Breast Cancer

摘要 目的初步探讨RABEX-5和RAB5与乳腺癌的关系。方法采用免疫组织化学法、RT-PCR和Western blot法检测RABEX-5及RAB5 mRNA和蛋白在60例乳腺癌、15例乳腺纤维腺瘤和15例乳腺正常组织中的表达,并检测相关临床病理指标与RABEX-5及RAB5表达的关系。结果 RABEX-5和RAB5 mRNA及蛋白在乳腺癌组织中的表达高于乳腺正常组织及乳腺纤维腺瘤(P<0.05)。在乳腺癌患者中,肿瘤直径(T)>5 cm组中的表达高于T≤2 cm组和2 cm<T≤5 cm组(P<0.05),在腋窝淋巴结转移阳性者中表达高于阴性者(P<0.05),但与乳腺癌组织学分级、人表皮生长因子受体2(HER-2)、雌激素受体(ER)和孕激素受体(PR)的表达无关(P>0.05)。RABEX-5和RAB5 mRNA及蛋白表达呈正相关(P<0.05)。结论 RABEX-5可能与RAB5共同参与了乳腺癌的发生及发展。 Objective To study the expression and significance of RABEX-5 and RAB5 in human breast cancer.Methods In this study the expression of RABEX-5 and RAB5 mRNA and protein was detected by Immunohistochemical tehniques,the reverse transcriptase polymerase chain reaction(RT-PCR) and Western blot assay in 60 cases of breast cancer,15 cases of breast fibroadenomas and 15 cases of normal breast tissue.The relationship of RABEX-5,RAB5 and the clinic pathological features was also analyzed.Results Expression of RABEX-5 and RAB5 in breast cancer was obviously higher than that in breast fibroadenomas and normal breast tissue(P〈0.05).RABEX-5 and RAB5 expression level in tumor diameter(T)〉5 cm group of human breast cancer was obviously higher than that in T≤2 cm and 2T≤5 cm group(P〈0.05).RABEX-5 and RAB5 expression level in axillary lymph node positive group was obviously higher than that in negative group(P〈0.05).However,there were no significant differences of RABEX-5 and RAB5 levels between HER-2(+) group and HER-2(-) group(P〉0.05),ER(-) group and ER(+) group(P〉0.05),PR(-) group and PR(+) group(P〉0.05),and in histological grade Ⅰ,Ⅱ and Ⅲ,there also were no significant differences of RABEX-5 and RAB5 levels.The expression of RABEX-5 is correlated with RAB5 in both mRNA and protein level.ConclusionRABEX-5 and RAB5 may be involved in the development of breast cancer metastasis.
