C反应蛋白基因型与慢性牙周炎和冠心病相关性研究 被引量:4

Relationship of C-reactive protein genetic polymorphisms with chronic periodontitis and coronary heart disease

摘要 目的:探讨CRP基因(+1059G>C rs1800947,+1444C>T rs1130864)的基因型、单倍型与慢性牙周炎、冠心病、慢性牙周炎伴冠心病易感性的关系。方法:中重度慢性牙周炎患者100例(CP组)、冠心病患者98例(CHD组)、中重度慢性牙周炎伴冠心病患者92例(CP伴CHD组)与125例健康对照者(对照组)的颊黏膜拭子,提取DNA,采用聚合酶链反应-限制性片段长度多态性法对+1059G>C rs1800947,+1444C>T rs1130864位点的基因分型,及对PC伴CHD组和对照组测序检测2个多态性位点单倍型。结果:CRP多态性位点rs1800947和rs1130864的等位基因在各组间差异无统计学意义(P>0.05)。2个多态性位点的等位基因在CP伴CHD组差异有扩大的趋势。CRP基因位点(rs1800947,rs1130864)的单倍型GT在CP伴CHD组和对照组间比较差异有统计学意义(P<0.05)。结论:单倍型GT可能是冠心病与慢性牙周炎的易感基因。 Objective To investigate the relationship of C-reactive protein polymorphisms(+1059GC rs1800947,+1444CT rs1130864) and haplotypes with the susceptible possibility of chronic periodontitis and/or coronary heart disease.Methods DNA was obtained from buccal swabs of 100 patients with moderate to severe chronic periodontitis(CP group),98 patients with coronary heart disease(CHD group),92 patients with moderate to severe chronic periodontitis with coronary heart disease(CP+CHD group) and 125 matched healthy controls(control group).The genetic polymorphisms of +1059GC rs1800947 and +1444CT rs1130864 were analyzed with PCR-PFLP technique and the genetic haplotypes were identified by sequencing in CP+CHD group.Results There was no significant difference in +1059GC rs1800947 and +1444CT rs1130864 distribution of genotype among the four groups(P0.05).The difference in +1444CT rs1130864 polymorphism had the tendency of expanding in CHD+CP group.There was a significant difference in haplotype GT of rs1800947 and rs1130864 between control group and CP+CHD group(P0.05).Conclusion The allele of haplotype GT is the predisposing gene of coronary heart disease and chronic periodontitis.
