补肾、活血组分及其复方制剂对着床障碍小鼠血管内皮生长因子的影响 被引量:6

The effect of bushen recipe and huoxue recipe on VEGF in mice of embryo implantation dysfunction

摘要 目的:比较补肾安胎方、补肾组分(补肾方)、活血组分(活血方)对胚泡着床障碍小鼠子宫内膜容受性的影响。方法:采用吲哚美辛诱导小鼠胚泡着床障碍模型,予补肾安胎方、补肾方、活血方治疗。于妊娠第5、6天处死小鼠,留取血清及子宫标本,采用免疫组织化学的方法检测VEGF蛋白的表达,采用RT-PCR的方法检测子宫VEGF mRNA的含量。结果:补肾安胎组小鼠子宫内膜中VEGF蛋白和mRNA含量均较模型组高;而补肾组和活血组VEGF蛋白及其mRNA均较模型组稍高,但差别无统计学意义;而补肾组于妊娠的第6天mRNA含量则与模型组相当。结论:补肾安胎方可以提高着床障碍小鼠子宫内膜VEGF蛋白和mRNA的含量,起到促进着床的作用,而补肾方和活血方单独使用不能显著提高小鼠子宫内膜VEGF蛋白和mRNA的含量。 Objective:To observe the effects of bushenantai recipe and its bushen and huoxue components(bushen recipe and huoxue recipe) on the endometrial receptivity in mice of embryo implantation dysfunction. Methods:Using the model of embryo implantation dysfunction induced by indomethacin in mice,to treate with bushenantai recipe,bushen recipe and huoxue recipe.The mice were killed on five and six days of pregnancy,their serum and uterine were collected.To detect the expression of VEGF protein by Radioimmunoassay and to detect the quantity of VEGF mRNA in uterus by RT-PCR. Results:The VEGF protein and mRNA in mice endometrial of bushenantai group were higher than the model group,but the VEGF protein and mRNA of bushen and huoxue group were all a little higher than the model group(except the bushen group of pregnant day 6),but there was no significant difference.Although the comparison of VEGF protein and mRNA of bushenantai group,bushen group,huoxue group were not exactly the same,in generally speaking,the VEGF protein and mRNA of bushen group,huoxue group in the endometrial were less than bushenantai group. Conclusion:Bushenantai recipe can improve the VEGF protein and mRNA levels in endometrial of implantation dysfunction mice,and also can play a role in the promotion of embryo implantation,but using bushen or huoxue recipe alone can not significantly improve the VEGF protein and mRNA levels in mice endometrial.
