摘要
目的:探讨马钱子配伍生地黄(以下简称马生)前后对大鼠佐剂性关节炎(AA)的治疗作用、相关机制及最佳配伍比例。方法:马钱子、生地黄不同比例水煎剂通过灌胃给药,采用Wistar大鼠复制AA模型,于给药1、8、12、16、20、24、28d测定各组大鼠足跖容积,计算免疫器官系数,采血并用硝酸还原酶法测定NO含量,羟胺法测定SOD的活力,硫代巴比妥酸(TBA)法测定MDA含量。结果:马生较单用马钱子对大鼠AA模型的关节肿胀度有明显抑制作用,其中以马生1∶6最佳,可使大鼠胸腺系数增大,脾系数减小,且可降低AA大鼠血清NO和MDA含量,升高SOD水平。结论:马生可防治AA,并对免疫系统有保护作用,其作用机制可能与抑制前炎症细胞因子生成、抗氧化等有关。其较佳配伍比例为马生1∶6。
Objective:Adopting the Adjuvants Arthritis(AA) rats to explore the treatment function,exploring mechanism and best compatibility proportion on the compatibility of Nux vomica and Dried rehmannia.Methods:Copying AA model and giving different compatibility proportion water decoction to the Wistar rats.Calculating the rats volume on first,eighth,twelfth,sixteenth,twentieth,twenty-fourth,twenty-eighth day and the coefficient of immune organs.Using the methods of nitrate reductase for NO determination,hydroxylamine for SOD determination,thiobarbituric acid(TBA) for MDA determination.Results:The compatibility had significant inhibitory effect on the joint swelling degree,could protect the immune system response in rats.At the same time,decreasing the contents of NO,MDA in blood serum and increasing the content of SOD,the results were superior to single decoction group.Conclusions:On one hand,the experimentation showes that the compatibility have the better anti-inflammatory and immune system balance effect,the mechanism is that it can lower lipid peroxidation and the restoration of antioxidant enzyme activity.On the other hand,it proves that the best proportion of the compatibility of Nux Vomica and Dried Rehmannia is 1∶6.
