摘要
目的:建立龙眼肉药材中果糖、葡萄糖、蔗糖的HPLC—ELSD含测方法,为科学评价龙眼肉质量提供依据。方法:采用Phenomenex Luna NH2柱(4.6mi/1×250into,5μm),乙腈-水(80:20)为流动相。结果:果糖、葡萄糖、蔗糖均能很好分离,线性范围和相关系数分别为:1.532~7.660μg(r=0.9998)、1.554—7.770μg(r=0.9999)及2.032~10.160μg(r=0.9998),平均回收率(n=9)分别为101.26%(RSD=1.87%)、101.39%(RSD=1.11%)及102.91%(RSD=1.19%)。结论:该方法简便、准确,分离效果好,无干扰,适用于该药材中糖类成分含量测定。
Objective: To establish an HPLC-ELSD method for the determination of fructose, glucose and sucrose in Arillus longan,and to investigate their contents in different places. Methods: The sample were separated on a Phenomenex Luna NH2 column (4. 6 mm× 250 mm,5μm) eluted with acetonitrile-water (80: 20) as mobile phase. Results: The methodological studies showed that the Fruc- tose, Glucose and Sucrose were separated. A good linearity was obtained with the correlation coefficients ranged from 1. 532 to 7. 660 μg ( r = 0. 999 8 ), from 1. 554 to 7. 770 μg( r = 0. 999 9) and from 2. 032 to 10. 160μg ( r = 0. 999 8), respectively. The average recov- eries of the three components(n =9) were 101.26% (RSD = 1.87% ) ,101.39% ( RSD = 1. 11% ) and 102. 91% ( RSD = 1.19% ), respectively. Conclusion:It was proved that the method with HPLC-ELSD was reliable, simple and well separated. It can be used as determination of saccharides in Arillus longan.
