摘要
目的通过研究河豚毒素不敏感型(tetrodotoxin-resistant,TTX-R)hNav1.8钠通道蛋白在三叉神经痛(trigeminal neuralgia,TN)患者痛支神经中的表达水平,探讨TN的发病机制。方法以12例原发性TN第三支痛非手术治疗无效行手术治疗患者的下牙槽神经、舌神经为研究对象,与制备的hNav1.8钠通道蛋白抗体进行免疫组化反应,并对两组免疫组化结果应用KS400图像分析系统测量其阳性颗粒面积百分比,并对两组数据进行统计学分析。结果 hNav1.8钠通道蛋白在T患者患支神经中均存在表达,且痛支神经中的表达多于非痛支神经(P<0.01),而正常下牙槽神经及肌肉组织中无表达。结论hNav1.8钠通道蛋白在TN患者痛支中的异常表达可能与TN的发病机制有密切关系。
Objective To study the pathogenesis of TN through assaying the expression of tetrodotoxin - resistant hNavl. 8 sodium channel protein in affected nerves of patients with TN. Methods Twelve affected inferior alveolar nerves and lingual nerves obtained from patients with idiopathic trigeminal neuralgia, to whom the drug therapy was not effective, were studied. These tissues and prepared hNavl. 8 antibody were examined by immunohistochemistry. The positive particle areas of the results of immunohistochemistry response were determined by KS400 photo analysis system and statistical analysis. Results The expression of hNavl. 8 channel protein was found in all of the 24 specimens of the affected nerves of patients with TN. Furthermore, the relative amount of hNav1. 8 sodium channel protein from the affected divisions was much higher than that from the non - affected divisions ( P 〈0.01 ). Conclusions The abnormal expression of hNavl, 8 channel protein in the affected nerves of patients with TN may play an important role in the pathogenesis of TN.
