摘要
以H(1967)为基本培养基、烟草花药为材料,研究了烟草花药培养中影响出苗率的因素。结果是:提高原H配方中微量元素的倍数或降低大量元素的倍数有利于烟草花培出苗;活性碳的促进作用与培养基中大量元素和微量元素之间的比例有关;活性碳的浓度以1.0%~2.0%出苗效果最好;活性碳经水洗后促进效果减弱,活性碳提取液有促进出苗的作用;活性碳杂质中Fe和Zn的作用是主要的。结果说明:活性碳在烟草花药培养中的作用主要是通过其杂质中Fe和Zn等增加了培养基中微量元素的含量,使培养基中微量元素与大量元素的比例趋于平衡,促进胚状体的形成和发育,提高出苗率。
