Foreign NPT Ⅱ gene was introduced into calli and mature embryos
of wheat strains 922 267, 942 114 and 953 1575 by using PDS 1000/He particle delivery
system. Plasmid pZFX 2 contains the NPT Ⅱ (neomycin phosphotransferase) and glycolate
oxidase gene which can increase the wheat plant resistance to insects. After selection culture
based on MS medium containing 100 μg/mL Kanamycin, resistant calli were selected. Green
plants were regenerated from part of resistant calli. Through this test, we found that the
optimum pressure of bombarding calli was 1 100 psi. Using this pressure, more resistant calli
were obtained. Mature embryos were bombared at pressure of 1 300 psi. It is worth to notice
that the green plants derived from transformed mature embryos were easier than those from
transformed calli.
