普通野生稻抗褐飞虱种质资源“94-42”的花药培养 被引量:3

摘要 采用N6诱导培养基和MS分化培养基,对普通野生稻(OryzarufipogonGrif)抗褐飞虱种质资源94-42进行了花药培养。愈伤组织诱导率为2.2%,绿苗分化率为12.4%,共获得18丛独立花药起源的绿苗共450株。在14丛共137株成活的花粉植株中,双单倍体占61.3%,单倍体占38.7%,其中3个独立起源的株系群共16个完全纯合株系对褐飞虱具有高度抗性,且抗性一致、无分离,抗性强度显著地高于任何一个已知的褐飞虱抗性基因所具有的抗性强度。 In vitro anther culture of 9442,a brown planthopper(BPH)resistant source of common wild rice Oryza rufipogon Griff was conducted,using N6 medium for callus induction and MS medium for plant regeneration.The frequencies of callus induction and plant regeneration were 22% and 124%,respectively.450 greenplants derived from 14 independent anthers were obtained.Of the 137 pollen plants survived after transplanting,613% were double haploids with normal fertility,while 387% were haploids.The selfed seeds harvested from the double haploids were used to identify homozygous BPH resistant lines.Sixteen lines derived from 3 independent anthers were completely homozygous and highly resistant to Pantnagar BPH biotype.These lines showed no segegation,and their BPH resistance was obviously higher than any known BPH resistant genes or sources from AA genome in rice.
