Toll样受体信号通路对大鼠创伤后深静脉血栓形成影响的实验研究

摘要 目的 探讨Toll样受体信号通路在创伤性深静脉血栓(TDVT)形成中的意义.方法 将30只SD大鼠随机分为正常对照组(A组)和模型组.模型组根据造模后的不同生物学状态再分为两组:高峰期血栓形成组(B组)、高峰期血栓不形成组(C组),在相应时相点无创切取股静脉血管组织,随后抽取各组大鼠总RNA,用Genechip Rat Genome 430 2.0芯片测定股静脉RNA表达,并分析Toll样受体信号通路基因表达的变化情况.结果 Toll样受体信号通路中TLR2、CD14、IFNα/βR、Rac1、TOLILP、PI3K、AKT、CASP8、IL-1β、IL-6、MIP-1α、IP-10、I-TAC等关键基因均上调,下调的有IL-12.结论 Toll样受体信号通路可能是调控血栓生物学状态的重要信号通路之一. Objective To study the significance of Toll - like receptors signaling pathway in traumatic deep vein thrombosis. Methods 30 SD rats were divided into control (group A) and experience groups. The experience groups were divided into 2 groups according to biological states: thrombosis at the crest - time ( group B) and without thrombosis at the crest - time ( group C). To incise the femoral vein in corresponding time, and to adopt to Trizol one - step method for total RNA extraction of femoral vein. After applying Genechip Rat genome 430 2. 0 genechips to detect the RNAs expressions, the genes related to Toll - like receptors signaling pathway were selected through geneehip data analysis. Results The key genes related to Toll -like receptors signaling pathway including TLR2, CD14, IFNα/βR, Racl, TOLILP, PI3K, AKT, CASP8, IL-1β, IL-6, MIP- 1α, IP- 10, I -TAC were up - regulation, and IL - 12 was down - regulation. Conclusion Toll - like receptors signaling pathway may be one of the most important pathway to control the biological states of thrombi.
