Quantitative Analysis of Plant Hormones With Capillary Electrophoresis

摘要 详细研究了三种重要植物激素,生长素吲哚-3-乙酸,吲哚-3-丁酸,赤霉素GA3,GA4及激动素6-苄氨基嘌呤的毛细管电泳分离条件;10min内完成上述5组分分离;分离效率为5×10^5 ̄2.6×10^6,5组分检出限为4.5 ̄18mg/L;迁移时间与峰高定量精度分别低于1.2%和2.4%,方法已用于香蕉叶中激素的测定。 In this study, it is reported the successful application of capillary electrophoresis for the separation of five plant hormones: indole-3-acetic acid(IAA), indole-3-butyric acid(IBA), 6-benzyl amino purine(6BAP), and two gibberellin( GA3 , GA4) . Under the optimum condition, the separation and. quantification of the above five plant hormones can be completed in 10 min, and their separationefficiency, plate number, are 5×105~2.6×106. With the length of 50 Um capillary, the detectionlimits(k=3) for IAA, IBA, 6BAP, GA3 and GA4 are 4.5×10-6 g/mL, 4.5×10-6 g/mL, 7.5×10-6g/mL, 18×10-6 g/mL and 18X10-6 g/mL, respectively. The analysis precision (n =11) is in 1.2%and 2. 4% for the measurement of the migration time and the determiantion with peak height, respectively. The approach has been used successfully to the analysis of plant hormone in banana leaf.
