摘要
目的观察人趋化素样因子1(CKLF1)对人外周动脉血管平滑肌细胞(ASMCs)的作用。方法用CKLF1真核细胞表达载体瞬时转染293T细胞,72 h后收集上清培养液,采用ELISA方法检测培养上清中CKLF1;用该上清培养人ASMCs细胞后,MTT法检测该上清对细胞增殖的影响。结果 MTT结果显示,10倍稀释的转染上清刺激ASMCs 72 h后,实验组OD490=0.480±0.024,与对照组OD490=0.424±0.011相比,差异有统计学意义(P<0.05)。结论 CKLF1对人ASMCs具有促增殖作用。CKLF1可能参与动脉粥样硬化血管内膜增厚的过程。
Objective To investigate the effect of human ehemokine-like factor 1 ( CKLF1 ) on proliferation of arterial smooth muscle cells (ASMCs). Methods The recombinant eukaryotic expression vectors pEGFP-N1-CKLF1 (test group) and pEGFP-N1 (control group) were transiently transfected into 293T cells. The supernatants were harvested 72 h after transfection for bioaetivity study. The effect of CKLF1 on ASMCs proliferation was evaluated by MTT. Results The 10 fold diluted supernants of CKLF1 transfected 293T cells promoted the perliferation of ASMCs. The absorbanee(490 nm) of test group had a statistical significance(stimulated with 72 h) (P 〈 0. 05 ) compared with that of control group. Conclusions CKLF1 has an effect on ASMCs proliferation. CKLFI may be involved in the vascular intimal thickening process of atherosclerotic.
