黄体酮对脑梗死大鼠缺血半暗带内凋亡诱导因子表达的影响

摘要 目的观察黄体酮对脑梗死大鼠缺血半暗带脑组织内凋亡诱导因子(apoptosis-inducing factor,AIF)表达及脑梗死体积的影响。方法选择成年雄性SD大鼠108只,体重260~310 g,并将其随机分为假手术组、模型组、溶剂治疗组、黄体酮治疗组。分别于脑梗死后12、24、48 h三个时间点处死大鼠,取出脑组织,通过免疫组化染色、Western印迹技术检测AIF的表达情况,并通过TTC染色检测脑梗死体积的变化。采用单因素方差分析对上述结果进行分析,检验水准α=0.05。结果脑梗死发生后,模型组及溶剂治疗组大鼠缺血半暗带内AIF的表达水平及脑梗死体积明显升高。经黄体酮治疗后,AIF的表达较模型组和溶剂治疗组有均有明显下降(P<0.05),脑梗死体积较模型组和溶剂治疗组亦明显缩小(P<0.05)。结论黄体酮可以明显抑制脑梗死大鼠脑组织内AIF的表达,并减轻梗死侧半球的脑损伤程度,具有明显的神经保护作用。
