摘要 目的观察姜黄素对人食管癌EC-9706细胞增殖的影响并探讨其分子机制。方法应用50~200txmol/L姜黄素处理食管癌EC-9706细胞株12—48h后,应用四甲基偶氮唑蓝(MTF)法检测细胞增殖抑制率、逆转录聚合酶链反应(RT—PCR)法检测热休克蛋白70(Hsrv0)mRNA的表达。结果经姜黄素干预后,人食管癌EC一9706细胞生长减慢,MTT法检测结果显示增殖抑制率达18.3%~77.5%(P〈0.05),呈良好的时间一剂量效应关系,HSP70mRNA的表达下调。结论姜黄素能够有效抑制人食管癌细胞EC-9706的增殖.其机制可能与抑制HSP70mRNA的表达有关。 Objective To explore the effect of curcumin on the proliferative inhibition of human esophageal cancer cell line EC -9706 and study the molecular mechanism. Methods After EC -9706 cells were treated with 50 -200 μmol/L curcumin for 12 -48 h, Methyl thiazolyl tetrazolium (MTF) method was used to detect the inhibition rates, and the expression of HSP70 mRNA was analyzed by RT- PCR. Re- suits After curcumin treatment, the growth of EC -9706 cells was inhibited significantly in a time and dose - dependent manner, and the inhibition rates were 18.3% - 77.5% ( P 〈 0.05). The expression of HSP70 mRNA was down - regulated ( P 〈 0.05 ). Conclusions Curcumin could effectively inhibit the proliferation of EC -9706 cells.Its mechanism may be inhibition of HSP70 mRNA expression.
