摘要
目的检测鸟氨酸脱羧酶(ODC)和c—myc在浅表性胃炎、萎缩性胃炎伴肠化生、不典型增生及胃癌组织中的mRNA表达水平,分析二者表达的差异性和相关性,探讨ODC及c—mye在慢性胃炎胃黏膜化生和癌变过程中的作用。方法应用逆转录一聚合酶链反应(RT—PCR)方法检测ODC和c—mycmRNA在18例浅表性胃炎、18例萎缩性胃炎伴肠化生、12例不典型增生及30例胃癌组织中表达,并做统计分析。结果ODCmRNA在萎缩性胃炎伴肠化生、不典型增生及胃癌组织中的表达水平显著高于在浅表性胃炎中的表达(P〈0.01)。此外,ODC在高分化腺癌的表达水平明显低于低、中分化腺癌(P〈0.01)。c—mycmRNA在胃癌组织中的表达水平显著高于在浅表性胃炎、萎缩性胃炎伴肠化生(P〈0.01),c-myc在低分化腺癌的表达水平明显高于高、中分化腺癌(P〈0.01)。ODC与c—myc在胃黏膜癌变多阶段中的表达呈正相关(P〈0.01)。结论ODC作为致癌因子在胃癌发生中有重要作用,且c—myc通过诱导ODC的表达促进细胞增殖,将两指标联合检测,有助于胃癌及其癌前病变的早期诊断。
Objective We detected the expression of ornithine decarboxylase and c -myc mRNA in gastric carcinoma and precancerous lesions, to analyse the differences and relevance of the two, and to investigate the role of ODC and c - myc in the process of the intestinal metaplasia as well as their role in gastric carcinogenesis. Methods The PT - PCR method was used to detect the expression of ODC and c - myc mRNA in 18 cases of chronic superficial gastritis(CSG) , 18 cases of chronic atrophic gastri- tis with intestinal metaplasia CAG(with IM),12 cases of gastric dysplasia (DYS) and 30 cases of gastric carcinoma (GC). Results The expression of ODC was lower in CSG than in CAG (with IM), DYS and GC( P 〈0.01 ). In addition,the expression of ODC was lower in well differentiated adenocarcinorma than in poorly and moderatedly differentiated adenocarcinorma ( P 〈 0.01 ). The expression of c - myc was higher in GC than in CSG and CAG (with IM) ( P 〈0.01 ). Besides, the expression of c -myc was higher in poorly differentiated adenocarcinorma than in well and moderatedly differentiated adenoearcinor- ma ( P 〈0.01 ). The expression of ODC mRNA was closely related to c - myc mRNA in gastric carcino- genesis ( P 〈 0.01 ). Conclusions ODC may play an important role as carcinogenic factor, and c -myc promotes cell proliferation by inducing ODC expression. Detecting both markers together may help in early diagnosis of gastric carcinoma.
