摘要
目的:观察黄柏对尿酸性肾病大鼠的影响及作用机制。方法:将SD雄性大鼠分为正常对照组、模型对照组、生理盐水组、黄柏治疗组、别嘌呤醇治疗组,酵母和腺嘌呤合用造模,造模成功后分别给予生理盐水、黄柏、别嘌呤醇灌胃,30 d后检测各组大鼠血中尿素氮(BUN)、肌酐(Cr)、尿酸(UA),并比较各组肾脏组织切片中环氧化酶2(COX-2),肿瘤坏死因子α(TNF-α)的表达。结果:治疗30 d后,与生理盐水组相比,黄柏治疗组大鼠血UA、BUN、Cr明显降低(P<0.05),COX-2与TNF-α表达明显比生理盐水组明显减少(P<0.05)。结论:黄柏对尿酸性肾病大鼠有明显的治疗作用,机制可能与其降低血尿酸,减少肾小管尿酸盐沉积及抑制相关炎性反应有关。
Objective: To observe the effect and mechanism of cortex phellodendron on uric acid nephropathy in rats.Methods: Male SD rats were divided into five groups including control group,model control group,sodium chloride group,cortex phellodendron treated group,and allopurinol treated group.Uric acid nephropathy rat models were established by using yeast and adenine,then rats were treated with Sodium chloride,cortex phellodendron,or allopurinol for 30 days.Blood urea nitrogen(BUN),creatinine(Cr),uric acid(UA) in blood and the expressions of cyclooxygenase 2(COX-2) and tumor necrosis factor alpha(TNF-α) in renal tissue were detected.Results: Compared with the sodium chloride group,the levels of BUN,Cr,and UA in blood and the expression of COX-2 and TNF-α in renal tissue in cortex phellodendron treated groups significantly decreased.Conclusion: Cortex phellodendron has a beneficial effect on uric acid nephropathy in rats.It could decrease the expression of COX-2 and TNF-α in renal tissue of uric acid nephropathy rats so as to alleviate the damage of kidney and protect renal function.
