血脑屏障氧糖剥夺体外模型大鼠脑微血管内皮细胞miRNA表达变化 被引量:3

Changes of miRNA expression profiles in cultured brain microvascular endothelium of rat with blood brain barrier for oxygen glucose deprivation

摘要 目的研究体外血脑屏障氧糖剥夺模型大鼠脑微血管内皮细胞miRNA表达变化。方法提取纯化大鼠脑微血管内皮细胞,建立血脑屏障氧糖剥夺模型,模拟在体缺血,在氧糖剥夺60min,应用基因芯片技术检测氧糖剥夺前后miRNA表达谱。利用靶基因软件对部分高表达的miRNA进行靶基因预测。结果共有278个miRNA表达,与正常对照组相比,氧糖剥夺60min,有47个较对照组具有统计学意义,其中25个上调,22个下调。miR-9、miR-103、miR-107和miR-290的靶基因是紧密连接相关蛋白claudin-5;miR-122a、miR-200b、miR-200c和miR-429的靶基因是紧密连接相关蛋白occludin;miR-23a、miR-23b、miR-185、miR-144、miR-101、miR-137和miR-335靶基因是紧密连接相关蛋白ZO-1。结论血脑屏障氧糖剥夺模型脑微血管内皮细胞miRNA表达差异可能在血脑屏障氧糖剥夺病理过程中起着重要作用。 Objective To investigate the changes of miRNA expression profiles in cultured brain microvascular endothelium(BMECs) of rat with blood brain barrier(BBB) for oxygen glucose deprivation(OGD) . Methods The BMECs were isolated and purified,a OGD BBB model in vitro was established and exposed to OGD for 60 min. The expression profiles of miRNA was examined by GeneChip. The target genes of the partly upregulated miRNA were predicted. Results 278 pieces of miRNAs were detected by GeneChip in BMECs,47 pieces of miRNAs were expressed significantly differentially,among which 25 pieces of miRNAs were upregulated and 22 pieces were downregulated in BMECs compared with control group. Claudin-5 was target gene of miR-9,miR-103,miR-107 and miR-290,occludin was target gene of miR-122a,miR-200b,miR-200c and miR-429,and ZO-1 was target gene of miR-23a,miR-23b,miR-185,miR-144,miR-101,miR-137 and miR-335. Conclusion Differential expressed miRNAs might play important roles in the pathological process of BBB model for OGD.
