摘要
目的:研究人肝癌低分化SMCC-7721细胞株,高分化HepG2细胞株和人正常肝细胞株LO2的高尔基体转运蛋白p115和巨噬细胞移动抑制因子(Macrophage migration inhibitoryfactor,MIF)的表达情况。方法:采用RT-PCR方法检测3组细胞株中p115和MIF的mRNA表达水平。用Western blot方法和细胞免疫组化方法检测3组细胞株中p115和MIF的蛋白表达情况。结果:SMCC-7721,HepG2的p115和MIF mRNA表达和蛋白表达水平明显增加且SMCC-7721表达更强,与对照组正常肝细胞株LO2的表达量比较有显著差异(P<0.05);细胞免疫组化方法结果显示:SMCC-7721,HepG2中p115在主要位于除高尔基体外的细胞质中,但在细胞质中HepG2着色相对较淡,而LO2中位于细胞核旁的高尔基体上。MIF主要分布于细胞质中,其在SMCC-7721、HepG2、LO23组细胞表达量呈明显减弱,统计学有显著差异(P<0.05)。结论:p115和MIF的含量在SMCC-7721、HepG2、LO2 3组细胞株中呈递减性变化。说明随着细胞恶性程度增加,p115和MIF表达增高,提示二者可能与肿瘤细胞的异常分化有关。
Objective: To observe and compare the expressions and cellular distributions of p115 and MIF in a normal human hepatic cell line(LO2) and different hepatocellular carcinoma cell lines(SMCC-7721,HepG2).Methods: The expressions of p115 and MIF proteins were measured using Western blot analysis.The mRNA levels of p115 and MIF were detected by reverse transcription polymerase chain reaction(RT-PCR).p115 and MIF expressions were further studied with immunocytochemistry techniques.Results: The results of RT-PCR and Western blot showed that the expressions of p115 and MIF of control group(LO2) were obviously weaker than that of those HepG2 and SMCC-7721;furthermore,in HepG2 the expressions were weaker than in SMCC-7721.The results of immunohistochemical cell staining of the p115 showed that yellow or brown granules were located in the cytoplasm in different hepatocellular carcinoma cells(SMCC-7721,HepG2),but in LO2 cells most of them were located on the Golgi complex.The cell staining for MIF was located in the cytoplasm of the normal hepatic cell line and in the different hepatocelluar carcinoma cells.Moreover,The expression of MIF in LO2 was weaker than that of HepG2,and that of HepG2 was weaker than that of SMCC-7721.Conclusion: The expressions of p115 and MIF in different hepatocellular carcinoma cells are dramatically higher than those in the normal human hepatic cell line,and p115 and MIF are strongly expressed in the SMCC-7721 cell line.The expressions and distributions of p115 and MIF are related to the stage of differentiation of cancer cells.
