接骨Ⅰ号治疗兔桡骨骨折中BMP-7表达的实验研究

BMP-7 expression at the treatment of radius fracture using "Jie Gu Yi Hao" in rabbit

目的:探讨中药接骨Ⅰ号治疗对兔桡骨骨折中骨形态发生蛋白-7(Bonemorphogeneticprotein-7,BMP-7)表达的影响。方法:选取56只健康成年新西兰大白兔,制作双侧桡骨3 mm骨缺损模型,随机分为实验组和对照组,每组28只。实验组给予中药接骨Ⅰ号外敷治疗,对照组不给予治疗。分别于术后5、7、9 d及2、3、4、6周处死实验动物,其中2、4、6周行X片及组织切片HE染色检查以观察骨折愈合情况,第5、7、9天和2、3周行免疫组化及RT-PCR以了解BMP-7在治疗过程中的变化。结果:X线片提示实验组骨痂出现较对照组早、数量多、出现骨性愈合时间较对照组早。HE切片提示实验组纤维连接出现较对照组早、成骨细胞和破骨细胞及骨痂量较对照组多、骨性愈合时间较对照组早。实验组RT-PCR灰度值、免疫组化平均光密度值(Mean optical density,MOD)与对照组行两样本均数比较的t检验、差别有统计学意义(P<0.05)。结论:接骨Ⅰ号通过促进BMP-7的表达而促进骨折的愈合。

Objective： To investigate the variation of bone morphogenetic protein-7（BMP-7）before and after ＂Jie Gu Yi Hao＂ treatment in the radius fracture of rabbit.Methods： 56 rabbits of New Zealand were randomly assigned into 2 groups,experimental group and control group.3 mm-long bone defect were taken in both radii of each rabbit.The experimental group were treated by ＂Jie Gu Yi Hao＂,while the control group was not treated.After 2,4,6 w,sacrifice the experimental animal.Then,assess the fracture healing through X-ray and HE.After 5,7,9 d and 2,3 w,observe the changes of BMP-7 through RT-PCP and immunohistochemistry.Results： X-ray shows： experimental group＇s bone callus appeared earlier and having more quantity bone callus.The experimental group＇s fibronectin and osteoblast occurs earlier than control group through HE,also have more callus than control group.Finally,do statistical analysis as the mean comparison of two samples＇ test into gray value of RT-PCR and MOD of immunohistochemistry.The results show that the difference is statistically significant.Conclusion： ＂Jie Gu Yi Hao＂ can promote fracture healing by improving the expression of BMP-7.
