摘要
目的:观察硝普钠能否促进静脉移植的大鼠骨髓间充质干细胞(Bone marrow mesenchymal stem cells,BMSCs)向溃疡性结肠炎(Ulcerativecolitis,UC)大鼠结肠的迁移。方法:分离培养4、’,6-二脒基-2-苯基吲哚盐酸(4’,6-diamidino-2-phenylindole,DAPI)荧光标记SD大鼠BMSCs。免疫复合法建立UC大鼠模型,1、2组UC大鼠均经尾静脉注射DAPI标记BMSCs的细胞悬液,2组细胞悬液还含0.2 mg/kg体重的硝普钠。移植后第7、14天各处死5只大鼠,取溃疡及非溃疡部位结肠做恒冷切片,荧光显微镜下分别计数各组DAPI标记BMSCs在溃疡及非溃疡部位结肠的细胞数。结果:移植第7、14天,2组结肠溃疡部位阳性细胞数均明显高于非溃疡部位(P<0.01);移植第7天,2组溃疡及非溃疡部位的阳性细胞数均高于1组(P<0.05)。结论:静脉移植BMSCs的同时给予硝普钠能促进BMSCs向结肠溃疡部位的迁移。
Objective:To investigate the effect of Sodium nitroprusside(SNP) in promoting the migration of vein transplanted bone marrow mesenchymal stem cells(BMSCs)to the colon of ulcerative colitis rat.Methods:BMSCs of SD rat were isolated,cultured and marked with DAPI.Ulcerative colitis(UC) rat was induced with immune-combined TNBS/ethanol.UC rats in group 1 and 2 was received caudal vein injection of DAPI labeled BMSCs cell suspension;but 0.2 mg Sodium nitroprusside/kg body weight was also injected in group 2.5 rats in each group were sacrificed on day 7 and 14 after injection.The colon of ulcerative or non-ulcerative parts were prepared as cryostat sections.The numbers of BMSCs both in colon of ulcerative or non-ulcerative parts were counted under fluorescent microscope respectively.Results:On the 7th and 14th day after injection,the number of marked BMSCs in the colonic ulcerative part was more than that in non-ulcerative parts(P 0.01) both in group 1 and 2;On the 7th day,the marked BMSCs number of group 2 both in the colon of ulcerative or non-ulcerative parts were higher than those in group 1(P0.05).Conclusion:SNP could encourage vein transplanted BMSCs migranting to the injured colonic tissue by the way that vein transplantion of BMSCs combined supply of SNP at the same time.
