不同剂量白藜芦醇对星形胶质细胞Aβ损伤的影响

Effects of resveratrol on β-amyloid-induced injury in astrocytes

摘要 目的研究白藜芦醇对β淀粉样蛋白诱导星形胶质细胞的超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)及丙二醛(MDA)释放的影响和其对氧化损伤的保护作用,从而探索其对中枢神经保护作用的可能机制。方法采用不同浓度的白藜芦醇预处理星形胶质细胞12h,继之用25μmol/L浓度的β淀粉样蛋白进行损伤,然后检测细胞SOD、GSH-PX及MDA释放的含量,并对数据进行统计学分析。结果β淀粉样蛋白损伤后,细胞SOD、GSH-PX的释放量明显下降(P<0.05),MDA的释放量升高(P<0.05),而用1、10、100μmol/L白藜芦醇预处理则能升高损伤细胞的GSH-PX的释放量(P<0.05),并能降低损伤细胞MDA的释放量(P<0.05);10、100μmol/L白藜芦醇预处理则能升高损伤细胞的SOD的释放量(P<0.05)。结论星形胶质细胞受到β淀粉样蛋白损伤后,白藜芦醇能增加SOD和GSH-PX的释放量,降低MDA的释放量,提示白藜芦醇对星形胶质细胞具有保护作用。 Objective To study the effects of resveratrol on β-amyloid-induced superoxide dismutase(SOD),glutathione peroxide(GSH-PX) and MDA in cultured astrocytes and the protective effects of resveratrol on the central nervous system.Methods Astrocytes were treated with various concentrations of resveratrol for 12 h and then exposed to Aβ.Cell viability was measured by CCK-8 assay.The levels of SOD,GSH-PX and MDA were examined after injury.Results After treatment with resveratrol(1,10 and 100 μmol/L),the MDA intensity was decreased(P0.05).At the concentrations of 10 and 100 μmol/L,resveratrol elevated the levels of SOD and GSH-PX in Aβ injuried astrocytes(P0.05).Conclusion Resveratrol may have cytoprotective effect through the increase of SOD and GSH-PX,and the decrease of MDA.
