HINT1蛋白及其在肿瘤抑制中的研究进展

摘要 与脆性组氨酸三联体(fragile histidine triad FHIT)蛋白类似,同属于组氨酸三聚体(histidinetriad HIT)家族的组氨酸三聚体核苷结合蛋白1(histidine triad nucleotide-binding protein1 HINT1)蛋白,自2003年通过HINT1基因敲除小鼠模型发现HINT1蛋白可能发挥肿瘤抑制作用,随着研究的不断进展,越来越多的证据表明HINT1蛋白在调控基因转录通路中具有抑制作用:与CDK7、转录因子Mi、USF、Pontin/Reptin/β-catenin/TCF4复合物、AP1相互作用并抑制其活性,从而发挥肿瘤抑制作用。本文就HINT1蛋白的基因、结构、组织细胞分布、功能及在各种肿瘤中发挥肿瘤抑制作用的机制做一综述。
