摘要
目的了解苯酚(PH)对中国仓鼠肺成纤维细胞(V79细胞)的细胞毒性以及DNA损伤。方法采用甲基四唑蓝(MTT)法检测不同浓度(25、50、100、200、400 mg/L)苯酚在不同时间染毒(12、24、48h)后对V79细胞的毒性;采用单细胞凝胶电泳技术(SCGE)检测不同浓度(5、10、20、40、80 mg/L)苯酚在不同时间(12、244、8 h)致V79细胞DNA的损伤情况。结果苯酚对V79细胞生长产生抑制作用,在各浓度和时间组内细胞存活率均随着染毒浓度增大和时间的延长而降低。在25、200、400 mg/L组间比较,差异具有统计学意义(P<0.05);各时间组间比较,差异有统计学意义(P<0.05)。单细胞凝胶电泳结果显示,在设置的5个浓度范围内,彗星细胞拖尾率、彗尾DNA%、尾长和OLIVE尾矩随浓度增加而增加,阴性组、低浓度和较高浓度组之间差异有统计学意义(P<0.05)。结论在本实验条件下,苯酚能抑制V79细胞的增殖,苯酚的细胞毒性存在明显剂量-效应关系和时间-效应关系,并能引起DNA损伤,主要为单链断裂。
Objective To investigate the cell cytotoxicity and DNA damage of Chinese hamster lung fibroblast cells(V79 cells) induced by phenol in vitro.Methods V79 cells were treated with suspension of phenol at concentrations of 25,50,100,200 and 400 mg/L for 12,24,48 h.SCGE was used to detect the DNA damage of V79 cells after exposed to different concentrations(5,10,20,40,and 80 mg/L)of phenol for 12,24 and 48 h.Results As the concentration increased during any definite time,viability of the treated cells turned lower than that of control group.Simultaneously,at any concentration of phenol,the longer the exposure was,the lower would be the viability rate of V79 cells.There was statistics difference in 12,24,48 h groups and also in 25,200,400 mg/L groups.The SCGE results showed that the comet cells ratio,tail DNA%,tail length and olive tail moment increased as the concentrations increased.The statistic differences among the control group,lower concentration groups and higher groups were significant(P0.05).Conclusions Under this experiment condition,phenol has toxicity on V79cells and shows dose-dependent and time-dependent relationship.Phenol can also induce DNA damage,mainly the single-strand break(SSB).
