摘要
目的探讨IL-6基因-572C/G多态性与小儿全身炎症反应综合征(SIRS)的关系。方法选用病例-对照的研究方法,以本院53例SIRS患儿为SIRS组,并随机挑选50例健康儿童为健康对照组(所有儿童均为汉族,来自广东省深圳市),采用酚/氯仿法提取2组儿童外周血白细胞DNA,并制备血清。自行设计引物,应用聚合酶链反应对2组研究对象的IL-6基因启动子-572位点的特异性片段进行扩增,再应用限制性片段长度多态性法对扩增的特异性片段用限制性内切酶进行酶切。酶切产物采用琼脂糖凝胶电泳法进行检测,分析2组儿童IL-6基因-572位点基因型及等位基因频率的分布。应用酶联免疫吸附法(ELISA)检测2组儿童血清IL-6水平,观察不同基因型对其血清IL-6水平的影响。采用SPSS 13.0软件进行数据分析。结果 IL-6基因-572位点基因型和等位基因频率在2组间分布差异均有统计学意义(Pa〈0.05);SIRS组GG基因型和G等位基因频率均显著高于健康对照组(Pa〈0.05);携带G等位基因个体患SIRS的风险约是C等位基因型个体的5.91倍(95%CI为2.76~12.64);SIRS组血清IL-6水平显著高于健康对照组(P〈0.05);G等位基因携带者血清IL-6水平显著高于CC基因携带者(P〈0.05)。结论 IL-6基因-572C/G多态性与中国汉族儿童SIRS发病密切相关;血清IL-6水平可能受其基因多态性的影响。
Objective To investigate the relationship between interleukin-6(IL-6) gene-572C/G polymorphism and systemic inflammatory response syndrome(SIRS) in children.MethodsThe research design was a case-control study.A total of 53 children admitted to our hospital with SIRS served as SIRS group while 50 healthy children as healthy control group(All children were Han nationality of Shenzhen city,Guangdong province).DNA was extracted from peripheral vein blood by phenol / chloroform method and serum was also prepared.Primer DNA was designed to amplify PCR product covering the part of IL-6 gene promoter carrying the analyzed transversion.Then,this PCR product was cut with restriction enzyme by polymerase chain reaction-restriction fragment length polymorphism method.Cleavage product was detected by agarose gel electrophoresis.Genotype frequencies and allele frequencies of the children in 2 groups were analyzed.The serum level of IL-6 was measured by enzyme-linked immunosorbent assay.The data was analyzed by SPSS 13.0 software.ResultsThe frequencies of the genotype GG and G allele of the IL-6(-572) site in the SIRS group were significantly higher than those in healthy control group(Pa0.05).The risk of SIRS of the G allele carriers was 5.91 times that of the C allele carriers(95%CI: 2.76-12.64).The serum level of IL-6 in SIRS group was significantly higher than that in healthy control group(P0.05),the serum level of IL-6 with the G allele carriers was significantly higher than that with the CC gene carriers(P0.05).ConclusionsIL-6 gene-572C /G polymorphism is significantly correlated with the SIRS in Chinese Han children.Serum IL-6 level maybe affected by the gene polymorphism.J Appl Clin Pediatr,2011,26(6): 415-417
