摘要
利用荧光标记AFLP技术研究剑麻种质遗传多样性及亲缘关系。筛选8个多态性高、分辨力强的Pst I/Mse I引物组合对40份供试材料的基因组DNA扩增,共获得条带数785条,多态性条带数507条,扩增效率64.58%,平均每对引物扩增条带数98.13条。其中,以P-GAC/M-CAC引物的扩增效率最高,达到78.63%;其次是P-GTG/M-CTC,达到71.28%;扩增效率最低的引物是P-GTA/M-CTT,为52.94%。表明供试材料在DNA水平上,酶切位点的分布存在广泛的变异。各供试材料相似系数介于0.44~0.83,在相似系数0.51处可将40份种质划分为4大类。8组引物在40份供试材料中检测到数目不等的特异带型,这些特异带型对供试剑麻种质具有一定的鉴别价值。
The genetic diversity and relationship of sisal were analyzed by fluorescent AFLP technology.AFLP fingerprinting of 40 accessions with eight pairs of Pst I/Mse I primers revealed a total number of 785 unambiguous bands including 507 polymorphic bands,and the polymorphism frequency was 64.58%.Each pair of primers produced 98.13 polymorphic bands on average.The primer P-GAC/M-CAC had the highest polymorphism frequency(78.63%),followed by the primer P-GTG/M-CTC(71.28%).The primer P-GTA/M-CTT had the least polymorphism frequency(52.94%).These show there are abundant diversities about enzyme digestion sites in the accessions.The similarity coefficient of the 40 accessions ranged from 0.44 to 0.83.The 40 accessions were divided into 4 groups at the similarity coefficient of 0.51.Moreover,some specific bands were detected from the 40 accessions by using 8 pairs of primers,and could be used to distinguish the accessions under test.
