摘要
利用ISSR分子标记技术,对来自全国13个省(市、区)的60份假俭草种质进行了遗传多样性分析。从60个ISSR引物中选用13个多态性高、分辨率强的引物分别对供试材料的基因组DNA进行扩增,获得104条清晰可辨的条带,共有多态性带96条,多态位点百分率为92.31%。表明,供试品种在DNA水平上存在着广泛的变异。利用NTSYSpc2.10e软件进行检测表明,60份资源所检出的位点平均有效等位基因数、平均多样度和平均Shannon信息指数,分别为(1.468±0.129)、(0.277±0.064)和(0.425±0.085)。遗传相似系数介于0.569 1~0.910 6之间,平均为0.739 6;UPGMA法将60份种质分成2个类群。
Genetic diversity of 60 Eremochloa ophiuroides(Munro)Hackel from 13 different areas was revealed by ISSR-PCR(Inter-simple sequence repeat).A total number of 104 unambiguous bands were amplified from the 60 Eremochloa ophiuroides cultivars with 13 primers screened from 60 ISSR primers,96 of which were polymorphic,and their polymorphism frequency was 92.31%.As analyzed by NTSYSpc2.10e,the average value of effective number of alleles,Nei’s genetil diversity and Shannon information index were(1.468±0.129),(0.277±0.064),(0.425±0.085),respectively.The Nei′s genetic distances of the 60 E.ophiuroides cultivars ranged from 0.5691~0.9106,with a mean of 0.739 6.The tested E.ophiuroides cultivars were divided into 2 groups by UPGMA based on Nei′s genetic distances.
