二氢青蒿素对结肠癌细胞HCT116凋亡基因表达图谱的影响 被引量:4

摘要 目的探讨二氢青蒿素(DHA)诱导结肠癌细胞HCT116凋亡基因差异表达。方法利用RT ProfilerTM PCR ArrayHuman Apoptosis和实时定量PCR方法对DHA组和对照组HCT116细胞的84种凋亡基因表达进行mRNA水平的检测。结果与对照组比较,DHA组细胞中,35种凋亡基因表达水平显著改变,TNFα、TRAILR、CASP、GADD45等促凋亡基因的表达显著上调(Fold Up-regulation>2);BCL2、AKT、BAG1等抗凋亡基因表达显著下调(Fold Down-regulation<-2)。结论 DHA通过涉及内质网应激、死亡受体介导和线粒体途径等复杂的分子机制诱导HCT116细胞凋亡。 Objective To explore dihydroartemisinin(DHA)-induced changes in the expressions of apoptotic genes in HCT116 cells.Methods The expression levels of 84 apoptotic genes in HCT116 cells with or without DHA treatment were detected using RT ProfilerTM PCR Array Human Apoptosis and quantitative real-time PCR.Results In DHA-treated cells,the expression levels of 35 apoptotic genes changed significantly.Compared with controls,the expressions of some apoptotic genes such as TNFα,TRAILR,CASP,and GADD45 were up-regulated,while the expressions of some survival genes such as BCL2,AKT,and BAG1 were down-regulated.Conclusion DHA induces the apoptosis of HCT116 cells via a complicate mechanism involving endoplasmic reticulum stress,death receptor-mediated pathway,and mitochondrial pathway.
