三七总皂苷对免疫性肝损伤小鼠的保护作用 被引量:16

Protective Effect of Saponins of Panax notognseng on Immunological Liver Injury in Mice

摘要 目的:观察三七总皂苷(saponins of Panax notognseng,PNS)对卡介苗(bacillus calmette guein,BCG)联合脂多糖(lipoxygenase,LPS)所致免疫性肝损伤小鼠的保护作用。方法:昆明种小鼠随机分为正常组、模型组、PNS低剂量组(100 mg.kg-1)、PNS中剂量组(200 mg.kg-1)、PNS高剂量组(400 mg.kg-1)组、联苯双酯(150 mg.kg-1)组。采用BCG 2.5 mg/只iv致敏小鼠,第10天给予LPS 7.5μg/只iv攻击,复制小鼠免疫性肝损伤模型,连续给药10 d。分光光度法测定血清中丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST),肝匀浆中超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)含量;HE染色法观察肝组织病理学变化;RT-PCR技术观察肝组织肿瘤坏死因子(tumor necrosis factor,TNF)-αmRNA的表达情况。结果:PNS能降低免疫性肝损伤小鼠脾脏、肝脏指数,ALT,AST,MDA含量(P<0.05或P<0.01),抑制TNF-αmRNA表达,减轻肝组织病理损伤程度,升高SOD,GSH-Px活性(P<0.05或P<0.01)。结论:PNS对免疫性肝损伤小鼠具有一定的保护作用。 Objective: To observe the protective action of Panax notoginseng saponins(PNS) on immunological liver injury in mice.Method:Mice were randomly divided into five groups,control group,model group,low-dose(100 mg.kg-1) of PNS,middle-dose(200 mg.kg-1) of PNS,high-dose(400 mg.kg-1) of PNS and bifendate(150 mg.kg-1) group.The model of mouse with immunological liver injury was induced by injection of BCG and LPS from tail vein.The ALT,AST,MDA content,SOD and GSH-Px activities were assayed by spectrophotometry,hepatic tissues stained with hematoxylin and eosin(HE) were prepared for histological analysis to liver injury.Moreover TNF-α mRNA expression in liver tissue was detected by RT-PCR technology.Result:PNS could decrease the enlarged liver,spleen index,ALT,AST and MDA content(P 0.05 or P 0.01),prevented the reduction of SOD and GSH-Px activities in liver homogenate(P 0.05 or P 0.01).Meanwhile,the pathological injury of hepatic tissues was alleviated and the expression of TNF-α mRNA in liver tissue was inhibited.Conclusion:PNS showed significant protective action on immunological liver injury in mice.
