用琼脂糖包埋——小室电洗脱纯化DNA的新方法

Agarose-embedment and Electroelution is a New Method for DNA Purification

先将５５℃保温的ＤＮＡ粗提液与１．２％琼脂糖凝胶等体积混合，然后把混合物注入由１．５ｍｌＥｐｐｅｎｄｏｒｆ离心管制作的小室电洗脱装置进行电洗脱纯化。纯化后的ＤＮＡ能被限制性内切酶完全酶解，其纯度足以进行Ｓｏｕｔｈｅｒｎ杂交等分析。该法具有所需条件简单、操作方便且花费低等特点，可作为常规的ＤＮＡ纯化方法。

Porphyra yezoensis cellls were prepared from sea snail enzyme digests of thalli and total DNA was isolated from P.yezoensis cells lysed with SDS-Proteinase K solution. The crude extracts of total DNA were purified with agarose-embedment and electroelution method.In this method,crude DNA was embeded in agarose gel and eluted through electrophoresis to remove polysaccharides,the resulting DNA was of sufficient quality for digestion of restriction endonucleases. DNA bands were clearly observed in the restriction endonuclease patterns of P.yezoensis total DNAs. The presence of DNA bands in the restriction pattern of total DNA indicated that the genome of P.yezoensis may be small and simple.
