胰岛素对大鼠主动脉血红素氧合酶活性的影响 被引量:1

The effect of insulin on the heme oxygenase-carbon monoxide system in the basal and induced state in normal rat aorta

摘要 目的 观察胰岛素对Wistar鼠主动脉组织及体外培养主动脉平滑肌细胞基础和诱导状态血红素氧合酶 (HO)活性的影响 ,探讨胰岛素与血红素氧合酶 -一氧化碳系统 (HO -CO)以及血管舒缩功能的关系。方法 应用离体Wistar鼠主动脉且织孵育技术和主动脉平滑肌细胞培养技术 ,以胆红素生成量作为HO活性的指标 ,以培养上清液中碳氧血红蛋白 (COHb)含量作为内源性一氧化碳 (CO)产量的指标 ,从组织和细胞水平观察生理范围内不同浓度胰岛素对血管局部HO活性和内源性CO产生量的影响。结果  ( 1)基础状态下 ,梯度浓度胰岛素 4 0、80、160、3 2 0、64 0 μU/ml使主动脉HO活性分别增高 4 8 2 %、60 8%、80 7%、70 5 %、62 2 % (P <0 0 5或 0 0 1) ,使主动脉平滑肌细胞培养上清液中COHb产量分别增加 8 7%、18 6%、4 0 2 %、17 4 %、7 9% (P <0 0 5或 0 0 1)。 ( 2 )诱导状态下 ,胰岛素对主动脉组织HO活性及血和平滑肌细胞培养上清液中COHb产量均无影响。结论 胰岛素对血管局部HO -CO系统的影响与HO -CO系统的状态有关 ;在基础状态下 ,胰岛素可引起血管局部HO活性增强 ,在生理范围内呈浓度依赖关系 ,过高浓度的胰岛素对HO活性作用反而减弱 ;但在诱导状态下 ,胰岛素对HO活性并无明显影响。 Objective The aim of this study was to investigate the effect of insulin on the heme oxygenase-carbon monoxide(H0-CO)system in its basal and induced state.Methods (1)Isolated Wistar rat aorta was cultured in the media containing different concentrations of insulin 40 μu/ml, 80μU/ml, 160μU/ml, 320 μU/ml, 640 μU/ml and add heme 10 μmol/L(basal state) or 40μmol/L (induced state) for 24 hours. The relative activity of HO was measured with bilirubin (pmol/g/h). (2) The Wistar rat aortic smooth muscle cells were cultured in the solution containing insulin and heme (consistency same as above) for 24 hours too. The relative amounts of carbon monoxide released into the media were measured with the quantifying carbon monoxide hemoglobin (COHb). Results (1) The HO activities in the basal state were increased, by the insulin, 48.2%, 60.8%, 80.7%, 70.5% and 62.2%, respectively (P<0.05 or<0.01). (2) The COHb in the culture solution of VSMCs were increased, by the insulin (concentrations as above), 8.7%, 18.6%, 40.2%, 17.1%, 7.9%, respectively P<0.05 or <0.01). (3) In the induced state HO activity was not effected by insulin. Conclusion This study suggests that insulin can increase aorta HO activity in the basal state but not in the induced state, and the effect of insulin on HO-CO system is related to the state of HO.
